Title: Rediscovering Clathrin-Mediated Endocytosis of NMDA Receptors and Vascular Dementia---Discussion on the Protective Effect of Shenzhi Jiannao Prescription

Abstract

Introduction: To explore the effects of Shenzhi Jiannao prescription on clathrin-mediated endocytosis(CME)of NMDA receptors in Vascular Dementia (VD) rats, and rediscover the relationship between CME of NMDA receptors and VD. Methods: Vascular dementia model in rats was established by repeatedly clamping bilateral common carotid arteries and intraperitoneal injection of sodium nitroprusside. The rats were randomly divided into five groups: normal group, sham-operated group, model group, Shenzhi Jiannao group and Memantine Hydrochloride group. Behavioral tests of learning and memory ability were performed in each group after operation. HE staining and Nissl's staining were used to detect pathological changes. Western Blot and RT-PCR techniques were used to analyze the expression of Clathrin, NMDAR1 and Rab5b in hippocampal neurons of rats. Results: The learning and memory ability, the arrangement and morphology of neurons in hippocampal CA1 area and the expression of NMDA receptor in Shenzhi Jiannao group and Memantine Hydrochloride group were improved compared with model group. The expression of clathrin, Rab5b and cytoplasmic NMDAR1 were increased, while the expression of membrane NMDAR1 was decreased. Conclusion: The obstruction of CME of NMDA receptors leads to accumulation of excitotoxicity, which is one of the pathological mechanisms of VD. Shenzhi Jiannao Prescription can effectively improve the endocytosis process, reduce the toxicity of excitatory amino acids.
